Monoclonal Antibodies Bound to Subunits of the Integrin GPIIb-IIIa Are Internalized and Interfere With Filopodia Formation and Platelet Aggregation
نویسندگان
چکیده
The monoclonal antibodies Tab and AP3 are directed, respectively, against GPllb and GPllla, the subunits of the platelet fibrinogen receptor. When added together to platelets, these antibodies prevent adenosine diphosphate (ADPI-induced platelet aggregation, despite normal fibrinogen binding (Newman et al. Blood 69:668, 1987). To explore the cellular requirements of aggregation after fibrinogen binding, we used several techniques to study platelets treated with Tab and AP3, then stimulated with ADP. We used scanning and transmission electron microscopy to evaluate platelet morphology, immunolabelsurface replication to determine whether individual GPllbllla complexes clustered, immunocytochemistry on frozen thin sections to study the subcellular distribution of the integrin GPllb-llla and fibrinogen, and biochemical methods

متن کاملArg-Gly-Asp-dependent occupancy of GPIIb/IIIa by applaggin: evidence for internalization and cycling of a platelet integrin.
Using indirect immunofluorescence microscopy we examined the distribution and cycling of GPIIb/IIIa after binding to applaggin, a high-affinity Arg-Gly-Asp (RGD)--containing ligand. Resting, unfixed platelets were incubated with applaggin for 30 minutes at 37 degrees C, and bound applaggin was detected by an affinity-purified rabbit anti-applaggin antibody. Examination of intact cells showed a ...

We studied the dose-response effects of the F(ab')2 fragments of murine monoclonal antibodies to the platelet GPIIb/IIIa receptor (7E3 and 10E5) on in vivo platelet thrombus formation in a well-characterized monkey model in which the carotid artery is stenosed and thrombus formation is provoked and augmented by intimal damage and the infusion of subaggregating doses of epinephrine. Both antibod...
متن کاملIntegrin IIb 3-specific synthetic human monoclonal antibodies and HCDR3 peptides that potently inhibit platelet aggregation
The interaction of fibrinogen with integrin αΙΙbβ3 (GPIIb/IIIa), in part mediated by an RGD tripeptide motif, is an essential step in platelet aggregation. Based on their inhibition of platelet aggregation, three integrin αΙΙbβ3 inhibitors are clinically approved. The clinically most widely used integrin αΙΙbβ3 inhibitor abciximab is a chimeric mouse/human antibody that induces thrombocytopenia...
متن کاملQuantitation of membrane glycoprotein IIIa on intact human platelets using the monoclonal antibody, AP-3.
A murine monoclonal antibody specific for glycoprotein (GP)IIIa was prepared by immunization with a GPIIb- and GPIIIa-enriched Triton X-114 extract of platelet membranes. This antibody, designated AP-3, was shown by indirect immunoprecipitation to react solely with GPIIIa derived from either P1A1-positive or -negative individuals. The epitope on GPIIIa recognized by AP-3 is expressed on dissoci...
